Extended Spectrum Beta-Lactamase Production and Biofilm Formation in Salmonella Serovars Resistant to Antimicrobial Agents Rev. Bras. Ciênc. Avic.
Mandelli,JZA; Ehrhardt,A; Manto,L; Borges,KA; Furian,TQ; Weber,B; Rodrigues,LB; Santos,LR.
ABSTRACT Antimicrobial resistance is a serious public health problem and Salmonella spp. is highly resistant to antimicrobial agents. Biofilms are important in the food industry due to their formation on products, utensils, and surfaces and the difficulty in their removal. The objective of this study was to assess extended spectrum beta-lactamase (ESBL) production, antimicrobial resistance, and biofilm production of Salmonella isolated from poultry slaughterhouses. Antimicrobial susceptibility was assessed by the disk diffusion assay and ESBL by double diffusion disk assay using the beta-lactamase inhibitor (amoxicillin+clavulanate). The antimicrobials tested were: ampicillin, amoxicillin+clavulanate, aztreonam, ceftazidime, cefotaxime, chloramphenicol,...

Gárcia,Carlos M.B.; Sprenger,Lew K.; Ortiz,Efraín B.; Molento,Marcelo B..
This study aimed to report the presence of parasites resistant to the most used anthelmintic drugs in sheep in Colombia. Four farms (denominated farm 1, 2, 3 and 4) were selected where the animals were not treated with anthelmintics for two months before the trial. Animals with faecal egg count (FEC) above 150 and of different ages were allocated into six groups, each consisting of at least 5 animals. The drugs and dosages used were: ivermectin 1% (0.2 mg/kg), albendazole 25% (5 mg/kg), fenbendazole 10% (5 mg/kg), levamisole 10% (5 mg/kg), and moxidectin 1% (0.2 mg/kg). Anthelmintic efficacy was determined by the FEC reduction test (FECRT) with a second sampling 14 days post-treatment. The efficacy of albendazole and fenbendazole at farm 1 was above 95%,...

Gene trio signatures as molecular markers to predict response to doxorubicin cyclophosphamide neoadjuvant chemotherapy in breast cancerpatients BJMBR
Katayama,M.L.H.; Brentani,H.; Abreu,A.P.S.; Barbosa,E.M.; Oliveira,C.T.; Góes,J.C.S.; Brentani,M.M.; Folgueira,M.A.A.K..
In breast cancer patients submitted to neoadjuvant chemotherapy (4 cycles of doxorubicin and cyclophosphamide, AC), expression of groups of three genes (gene trio signatures) could distinguish responsive from non-responsive tumors, as demonstrated by cDNA microarray profiling in a previous study by our group. In the current study, we determined if the expression of the same genes would retain the predictive strength, when analyzed by a more accessible technique (real-time RT-PCR). We evaluated 28 samples already analyzed by cDNA microarray, as a technical validation procedure, and 14 tumors, as an independent biological validation set. All patients received neoadjuvant chemotherapy (4 AC). Among five trio combinations previously identified, defined by nine...

HIV-1 subtypes and mutations associated to antiretroviral drug resistance in human isolates from Central Brazil BJM
Cerqueira,Daniela Marreco; Ramalho,Eduardo Dias; Oliveira,Claudiner Pereira; Silva,Ruiter Roberto; Franchini,Miriam; Felipe,Maria Sueli Soares; Martins,Cláudia Renata Fernandes.
The detection of polymorphisms associated to HIV-1 drug-resistance and genetic subtypes is important for the control and treatment of HIV-1 disease. Drug pressure selects resistant variants that carry mutations in the viral reverse transcriptase (RT) and protease (PR) genes. For a contribution to the public health authorities in planning the availability of therapeutic treatment, we therefore described the genetic variability, the prevalence of mutations associated to drug resistance and the antiretroviral resistance profile in HIV-1 isolates from infected individuals in Central Brazil. Nineteen HIV-1 RNA samples from a Public Health Laboratory of the Federal District were reversely transcribed and cDNAs were amplified by nested PCR. One fragment of 297 bp...

Inhibition of focal adhesion kinase by antisense oligonucleotides enhances the sensitivity of breast cancer cells to camptothecins Biocell
Satoh,Taroh H.; Surmacz,Tatiana A.; Nyormoi,Okot; Whitacre,Cecilia M..
This study shows a strong association between cell attachment to substratum and activation of b1-integrin-signaling with resistance to the camptothecin derivative topotecan (TPT) in breast cancer cells. We propose a mechanistic-driven approach to sensitize the cells to camptothecins. ZR-75-1 anchorage-dependent breast cancer cell line, its derivative 9D3S suspension cells (9D3S-S), and 9D3S cells attached to fibronectin-coated plates (9D3S-A) were treated with TPT (1 mM) or CPT-11 (40 mM) for 48 h. Programmed cell death (PCD), as shown by poly(ADP-ribose) polymerase (PARP), pro-caspase-3 and pro-caspase-9 cleavage, was observed in 9D3S-S cells but not in ZR-75-1 or 9D3S-A cells. Because p125 focal adhesion kinase (FAK) is a transducer in the b1-integrin...

Prevalence of carbapenem resistant Pseudomonas aeruginosa and Acinetobacter baumannii in high complexity hospital BJID
Baumgart,Ana Milda Karsten; Molinari,Marcelo André; Silveira,Alessandro Conrado de Oliveira.
Pseudomonas aeruginosa and Acinetobacter baumannii are Gram-negative bacilli that in the last decades have become prevalent agents of hospital infection due to high antimicrobial resistance developed by these microorganisms. The present study is a retrospective analysis of all positive cultures for these microorganisms in the period of January 2004 to December 2008. Resistance levels of A. baumannii and P. aeruginosa to carbapenems was high and showed a trend to increase during the period of study. In recent years the increasing incidence and resistance levels of A. baumannii and P. aeruginosa to the antimicrobials used for their treatment in the hospital setting underscores the relevance of infections caused by these bacteria. The selective pressure...

Probiotic and formaldehyde treatments of Artemia nauplii as food for larval pollack, Pollachius pollachius ArchiMer
Gatesoupe, Joel.
Formaldehyde was used to disinfect Artemia cysts and nauplii, while introducing two probiotics in the enrichment process: Bactocell (Pediococcus acidilactici) and Levucell (Saccharomyces cerevisiae). The disinfectant was selected due to its potential compatibility with probiotics, since it was more effective against Gram-negative bacteria than against lactic acid bacteria and yeast. However, the presence of formaldehyde reduced the intake of P. acidilactici in Artemia. Consequently, the disinfection was stopped before Bactocell supplementation to the nauplii fed to pollack (Pollachius pollachius) larvae. The mean weight of pollack was higher with this probiotic treatment. Growth was even better with the combination of Levucell and Bactocell, but the yeast...

Salinomycin enhances doxorubicin sensitivity through reversing the epithelial-mesenchymal transition of cholangiocarcinoma cells by regulating ARK5 BJMBR
Yu,Z.; Cheng,H.; Zhu,H.; Cao,M.; Lu,C.; Bao,S.; Pan,Y.; Li,Y..
Chemotherapy response rates in patients with cholangiocarcinoma remain low, primarily due to the development of drug resistance. Epithelial-mesenchymal transition (EMT) of cancer cells is widely accepted to be important for metastasis and progression, but it has also been linked to the development of chemoresistance. Salinomycin (an antibiotic) has shown some potential as a chemotherapeutic agent as it selectively kills cancer stem cells, and has been hypothesized to block the EMT process. In this study, we investigated whether salinomycin could reverse the chemoresistance of cholangiocarcinoma cells to the chemotherapy drug doxorubicin. We found that combined salinomycin with doxorubicin treatment resulted in a significant decrease in cell viability...

Synergistic effects of sulbactam in multi-drug-resistant Acinetobacter baumannii BJM
Temocin,Fatih; Erdinc,Fatma Sebnem; Tulek,Necla; Demirelli,Meryem; Ertem,Gunay; Kinikli,Sami; Koksal,Eda.
Abstract Acinetobacter baumannii is a frequently isolated etiologic agent of nosocomial infections, especially in intensive care units. With the increase in multi-drug resistance of A. baumannii isolates, finding appropriate treatment alternatives for infections caused by these bacteria has become more difficult, and available alternate treatments include the use of older antibiotics such as colistin or a combination of antibiotics. The current study aimed to evaluate the in vitro efficacy of various antibiotic combinations against multi-drug resistant A. baumannii strains. Thirty multi-drug and carbapenem resistant A. baumannii strains isolated at the Ankara Training and Research Hospital between June 2011 and June 2012 were used in the study. Antibiotic...
